In the presence of the target, Cas12a trans-cleaves the biotinylated ssDNA reporter, which results in short sequences that cannot bind a DNA probe pre-immobilized on the lateral flow biosensor (LFB) test line, resulting in an undetectable test line on the LFB. The control line appears due to the excess of gold nanoparticles-streptavidin (AuNPs-SA) conjugate complex captured by the biotin-labeled goat anti-mouse IgG immobilized on the control line.
